چکیده

Sir, Pseudomonas aeruginosa expresses between five and nine major outer membrane proteins (OMPs), depending on the conditions of growth; the genes encoding all but one of these, OprG, have been identified. OprG has an apparent molecular weight of 25,000 when outer membranes are heated to 88°C for 10 min in solubilization buffer before being subjected to SDS–PAGE and 19,000 when they are not pre-heated. Its appearance in the outer membrane is highly dependent on growth conditions. In particular, Yates et al. observed a direct relationship between the iron concentration in the medium and expression of OprG and suggested that this OMP is involved in low-affinity iron uptake. Other conditions, including growth into the stationary phase, higher growth temperatures, Mg deficiency, certain lipopolysaccharide alterations and the presence of certain carbon sources, also result in the expression of varying concentrations of OprG. Such apparently broad regulation of this OMP has frustrated efforts to assign a function to it. However, in addition to the potential role of OprG in iron uptake, two groups of investigators have suggested that it is involved in quinolone uptake/ susceptibility. In view of this uncertainty, we undertook to identify the oprG gene. Outer membranes were prepared according to the method described by Hancock & Carey from a 500 mL overnight culture of P. aeruginosa IA1 in L-broth (1% Bacto-Tryptone, 0.5% yeast extract; Difco Chemical Co., St Louis, MO, USA). The OMPs were electroblotted on to an Immobilon P polyvinylidinedifluoride membrane (Millipore, Bedford, MA, USA) which was then dried and stained. The OprG band was excised from the blot and subjected to N-terminal amino acid sequencing by Dr P. Williams (Department of Pharmaceutical Sciences, Univers i t y of Nottingham, Nottingham, UK). Outer Membrane Protein D Gene in Clinical Isolates of Pseudomonas Aeruginosa and its Role in Antibiotic Resistance

Background & Objectives: Pseudomonas aeruginosa is a common cause of nosocomial infection. OprD protein is a specific protein regulating the uptake of carbapenem antibiotic. Loss of OprD is the main mechanism of Pseudomonas Aeruginosa resistance to carbapenem. In this study, the presence of OprD gene is investigated in isolated Pseudomonas Aeruginosa in burn patients of Ghotboddin hospital in S...

The major outer membrane protein OprG of Pseudomonas aeruginosa contributes to cytotoxicity and forms an anaerobically regulated, cation-selective channel.

OprG of Pseudomonas aeruginosa is a member of the very large and widely distributed but poorly characterized OmpW (PF0392) family of outer membrane proteins. It was established here that OprG was highly transcribed in anaerobic environments rich in iron via the ANR regulator. In the absence of OprG, P. aeruginosa was significantly less cytotoxic toward human bronchial epithelial cells. Planar b...

The Crystal Structure of OprG from Pseudomonas aeruginosa, a Potential Channel for Transport of Hydrophobic Molecules across the Outer Membrane

BACKGROUND The outer membrane (OM) of Gram-negative bacteria provides a barrier to the passage of hydrophobic and hydrophilic compounds into the cell. The OM has embedded proteins that serve important functions in signal transduction and in the transport of molecules into the periplasm. The OmpW family of OM proteins, of which P. aeruginosa OprG is a member, is widespread in Gram-negative bacte...

Conserved OprF as a Selective Immunogen Against Pseudomonas aeruginosa

Background & Objectives: Due to the importance of Pseudomonas aeruginosa in severe inpatient infections and high mortality, the need for an efficient vaccine against these bacteria is increasing. In this regard, the general outer membrane porin of the most problematic microorganism P. aeruginosa, outer membrane protein F (OprF), is a good vaccine cand...

Influence of a putative ECF sigma factor on expression of the major outer membrane protein, OprF, in Pseudomonas aeruginosa and Pseudomonas fluorescens.

The gene encoding OprF, a major outer membrane protein in Pseudomonas species (formerly known as type 1 pseudomonads), was thought to be constitutively transcribed from a single sigma 70 promoter immediately upstream of the gene. We now report the identification of a novel putative ECF (extracytoplasmic function) sigma factor gene, sigX, located immediately upstream of oprF in both Pseudomonas ...
